One way car hire is possible between Amsterdam ferry port and Rotterdam. When you do your search tick the box "Return to a different location". When cars are shown after pressing the search button, cars are available based on your search criteria.

Yes, I know Amsterdam very well. From the port terminal you best take a cab to the Sixt office. It is about a 10 minutes ride but too far to walk. The Sixt office is located near the Amsterdam Central Train Station.
